Screened peptides and small molecules targeting CXCR4 to identify biased antagonists that inhibit G protein signalling but allow for beta-arrestin recruitment and avoid the development of tolerance.•Project: Discovery of small molecule antagonists of FOXM1. Using NMR and purified, recombinant FOXM1 DNA-binding domain, demonstrated that the small natural compound honokiol inhibits FOXM1 directly. Characterised the chemical epitope of the small molecule using 1D/proton NMR.•Project: Investigating the mechanism of PTK6 membrane localization and discovery of novel inhibitors. Using NMR, biophysical methods and recombinant PTK6, demonstrated that the PTK6 SH2 domain binds phosphatidylinositols to localize to the plasma membrane while the BRAF inhibitor PLX4032 can bind to the PTK6 catalytic domain to inhibit kinase activity. From September 2012 to March 2019 (6 years 7 months) Greater Chicago AreaResearch Scholar @ University of Illinois at Chicago Identified the subcellular localization of the small GTPase K-Ras4B to both loose and rigid plasma membrane microdomains. Worked with structural biologists to show the different mechanisms of interaction between K-Ras4B and different membrane domains.
